Digital PCR

In partnership with Stilla Technologies, Source Genomics offers digital PCR services using the advanced Nio instrument.

Digital PCR (dPCR) Services

Digital PCR (dPCR) is a highly precise and sensitive method of DNA and RNA quantification. Unlike traditional quantitative PCR (qPCR), which provides relative quantification, dPCR partitions a sample into thousands of tiny reactions, performing PCR amplification in each one separately. This allows for absolute quantification of nucleic acids without the need for standard curves.

How Digital PCR (dPCR) Works?

Partitioning:

The sample is divided into thousands of individual reactions (droplets, wells, or chambers) using Crystal Droplets.

PCR Amplification:

Each partition undergoes PCR independently.

Fluorescent Detection:

After amplification, partitions are analysed for fluorescence, identifying which contain the target DNA/RNA.

Absolute Quantification:

The fraction of positive partitions is used to calculate the absolute concentration of the target. This method uses end-point analysis and Poisson statistics to quantify the absolute number of nucleic acid molecules in a sample.

Key Benefits Of Digital PCR (dPCR):

Higher Sensitivity:

Detects low-abundance targets, even in complex samples.

Absolute Quantification:

No need for reference standards or calibration curves.

Better Precision & Reproducibility:

Ideal for rare mutation detection and copy number variation (CNV) analysis.

Resistant to Inhibitors:

Works well with challenging sample types.

Nio® Digital PCR:

An all-in-one, automation-friendly dPCR system with 7-color detection and capacity for up to 768 samples.

Nio® Digital PCR

Stilla is the only dPCR provider to have a system with 7 channels excitations (nm), which allows for a high level of multiplexing for targets, enabling up to 21 targets with the Crystal Flex probes.
